An activity-dependent assay for ricin and related RNA N-glycosidases based on electrochemiluminescence, , , , , a United States Army Medical Research Institute of Infectious Diseases, Frederick, MD, USAb BioVeris Corp., Gaithersburg, MD, USASynthetic biotinylated RNA substrates were cleaved by the combined actions of ricin holotoxin and a chemical agent, N,N&-dimethylethylenediamine. The annealing of the product with a ruthenylated oligodeoxynucleotide resulted in the capture of ruthenium chelate onto magnetic beads, enabling the electrochemiluminescence (ECL)-based detection of RNA N-glycosidase activities of toxins. ECL immunoassays and the activity assay exhibited similar limits of detection just below signals with 0.1 ng/ the ECL response was linear as the ricin concentration increased by two orders of magnitude. Activities were detected with other adenine-specific RNA N-glycosidases, including Ricinus communis agglutinin (RCA), saporin, and abrin II. The substrate that provided the greatest sensitivity was composed of a four-residue loop, GdAGA, in a hairpin structure. When the 2&-deoxyadenosine (dA) was substituted with adenosine (A), 2&-deoxyinosine, or 2&-deoxyuridine, toxin-dependent signals were abolished. Placing the GdAGA motif in a six-residue loop or replacing it with GdAdGA or GdAAA resulted in measurable activities and signal patterns that were reproducible for a given toxin. Data indicated that saporin and abrin II shared one pattern, while ricin and RCA shared a distinct pattern. Over the past twenty years the field of microbial infections has made spectacular progress with the recognition of the intense and sophisticated cross-talk between bacterial pathogens and the infected host. These advances result largely from appreciating that the properties of bacteria need to be studied in the context of their interaction with host cells. Cellular microbiology studies as they were defined by Cossart et al. (‘Cellular microbiology emerging’, Science 5–6) have yielded a wealth of information including the identification and extensive characterization of secretion systems, toxins, and adhesive structures. These cell-centered strategies allow the characterization of virulence factors at the molecular level with exquisite resolution in time and space. In parallel with these studies focused on the interaction with host cells, infectious diseases have been extensively studied in various animal models. Such studies have addressed global pathophysiological issues and are particularly efficient to follow disease progression and for identifying the sites of bacterial proliferation. Recent work have demonstrated the importance of combining these two levels of analysis to provide a detailed description of the infection process inside the animal, at the molecular level, and with high resolution.
In this issue of Current Opinions in Microbiology we have brought together scientists involved in cutting edge research on the molecular basis of host pathogens interaction in vivo, to share with us their view of this rapidly evolving field of research. We have chosen to limit the scope of the reviewsto mammalian infection models to obtain a concise view of this particular research field. We feel strongly that from this series of ‘current opinions’, an integrated, rich and exciting view of the mechanisms underlying infection, opening numerous novel avenues of research, clearly emerges. As indicated by the subtitle of this special issue, we here propose tissue microbiology as a suitable term to describe this emerging research field.
An increasing number of studies point to the importance of placing the infection in the context of the whole organism or at least in the context of the infected tissue. The fact that the infection occurs inside a whole organism has several consequences on the mechanisms underlying disease progression, which we have chosen to highlight in this issue. First, tissue architecture determines the infection process, there is tremendous diversity among the different organs, and organs are more than a juxtaposition of individual cells. Brain, blood vessels, lungs, urinary tact, gut, skin, and placenta are all well-studied infection sites with significant diversity. Throughout the organism the immune system shapes the infection process in a way that is specific to the infected organ. Depending on the site of infection pathogens need to adapt to different tissue-specific environmental factors. This includes the alteration of bacterial micro-environmental factors during progression of infection, as well as mechanical challenges, such as shear stress from vascular or renal filtrate flow. Flow induced shear stress is known to influence the molecular basis for bacterial binding to biotic and abiotic surfaces. Oxygen and iron availability represent important environmental factors that affect the infection process. Finally, bacterial infections usually occur in the presence of other bacterial species. Thus, not only is communication between microbes and host cells important, but also interbacterial and poly-microbial communication.
